7: Grappling with Goals and Gratitude

07/22/24 – Turning Up the Heat By Three Notches

Today marked a significant shift in lab intensity. Seeing David already working in the lab when I arrived underscored that the coming days would be challenging.

David and I tackled Western Blot optimizations, testing different antibody concentrations and conditions to refine our results. We focused on improving the TMEM109 and DCC antibodies, adjusting concentrations, and preparing new samples for more reliable data. Despite the behind-schedule feeling, we made progress by preparing new plates and setting up for upcoming experiments. A break to attend the NCI event revealed a lack of engagement from the organizers but allowed for a much-needed diversion and snack.

Back in the lab, we continued with various tasks, including preps for DOCK8 and MEA plate recordings. My focus shifted to refining the experimental processes and understanding the nuances of Western Blots. I also reflected on my role in the project, acknowledging both the hands-on experiences and the need to balance independence with team contributions.

07/23/24 – Searching for the Silver Lining

On this day, optimization took center stage as we addressed issues with DOCK8 and explored potential improvements. David suggested lowering the denaturation temperature to reduce band smearing, but we faced challenges due to limited samples. I shifted focus to optimizing the concentration of DCC, which better aligned with my previous research. Meanwhile, a conversation with Cristina provided valuable insights into international research and regenerative tissue bioprinting, highlighting Spain and the Netherlands as promising locations. Her advice on travel and research opportunities was a welcome addition to my growing understanding of the field.

David and I also tackled issues with our experimental designs, such as reworking our approach to the BCA assay and immunostaining. Conversations with Kody about the balance between hands-on and reading work helped me reassess my own contributions. Despite feeling disconnected from the project at times, I recognized the value of the iterative process in research and continued to refine my poster and figures. The day was a mix of technical challenges and personal growth, reinforcing the importance of adapting and learning through each experience.

07/24/24 – Gratitude for God is Good… And All the Time…

A renewed sense of gratitude transformed my outlook on the day’s challenges. I began by working on my poster figures with BioRender, while David handled the BCA assay and immunostaining preparations. Despite feeling isolated from my project, I focused on challenging my negative thoughts and appreciating the progress I had made. David’s compliments on my poster work and the opportunity presented by Mark from the 3D bioprinting lab were affirming. Reflecting on my journey, I recognized how my faith and experiences at NCATS had contributed to my growth, fostering a more positive and purposeful mindset.

Throughout the day, I grappled with feelings of inadequacy but found solace in understanding the significance of my contributions. As David and I prepared for the BCA assay and immunostaining, I worked on internalizing the lessons learned and appreciating the opportunities provided. Embracing gratitude allowed me to regain focus and energy, reaffirming the importance of maintaining a positive outlook amidst challenges. This shift in perspective highlighted the value of my experiences and the role of faith in guiding me through my research journey.

07/25/24 – Racing At Lightspeed

This day was defined by balancing a hectic schedule of coding homework, class commitments, and a looming poster deadline. The importance of bioinformatics became evident as I realized how essential it is for running tests and drawing conclusions from research data. Despite the stress of meeting the poster deadline, I approached the task enthusiastically and confidently, striving to convey my growth and understanding. A brief but impactful conversation with Jill Powell, the program director, bolstered my confidence and allowed me to express my ambitions clearly.

Completing the poster just minutes before the deadline was a significant achievement despite the challenges posed by my caffeine-induced cognitive fatigue. This day underscored the need for effective time management and the ability to handle multiple responsibilities simultaneously. The experience of finalizing the poster and preparing for my presentation was a valuable lesson in balancing stress with gratitude and maintaining focus on long-term goals. My commitment to managing tasks and staying positive was reinforced, paving the way for continued progress and growth.

07/26/24 – Regain Purposeful Living

With the pressure of deadlines behind me, my focus shifted to personal priorities and upcoming commitments, including the Ethiopian Evangelical Church Youth Legacy Conference. However, a lack of purpose and poor time management led to frustration, especially when I missed an opportunity to contribute effectively at the conference. Despite these setbacks, I recognized the importance of clearer communication about my needs and maintaining a balanced approach to work and personal life.

Reflecting on the day’s events, I realized that my efforts to fill time without a clear purpose ultimately hindered my progress. The experience highlighted the need for intentional planning and setting priorities. Embracing a more proactive approach to managing my time and responsibilities will be crucial in achieving my goals and maintaining a sense of purpose both in and outside the lab. This day reminded me of the importance of aligning my actions with my values and goals.
